In this study, the pharmacological effect of Phlomides isochila (IM-1) plant extract on isolated aortic smooth muscle was investigated. The research was aimed at evaluating the mechanisms of contraction and relaxation of vascular smooth muscle, which play a crucial role in the regulation of the cardiovascular system. Experimental studies were carried out on isolated aortic preparations, and the functional state of smooth muscle was assessed using classical pharmacological agonists. Contraction of aortic smooth muscle was induced by a high concentration of KCl. KCl causes membrane depolarization, leading to the opening of voltage-dependent calcium channels and the influx of Ca²⁺ ions into the cell. In addition, receptor-mediated contractions were induced by phenylephrine through α₁-adrenoceptors. These two experimental models made it possible to evaluate different physiological mechanisms of vascular smooth muscle activity. During the experiment, after stable contractions induced by KCl and phenylephrine were achieved, the IM-1 extract was applied at various concentrations. The obtained results demonstrated that the IM-1 extract caused a significant relaxation of aortic smooth muscle in both experimental models. This effect indicates that the extract may limit calcium ion influx, reduce smooth muscle tone, and exhibit vasorelaxant mechanisms of action. Thus, Phlomides isochila (IM-1) extract possesses strong vasorelaxant properties and may be considered a promising natural source for the development of new plant-based pharmaceutical agents for the treatment of cardiovascular diseases.
